摘要
目的:观测慢性缺氧和二氧化碳潴留大鼠脑桥组织中脑阴离子转运蛋白(AE3)mRNA的变化。方法:采用密闭舱内灌注混合气体的方法,分别复制慢性缺氧,慢性缺氧伴二氧化碳潴留及慢性呼吸性酸中毒大鼠模型,以地高辛标记的(AE3)cDNA探针,应用点杂交技术,对各实验组及对照组大鼠脑桥组织中AE3mRNA的相对含量进行测定。结果:(1)慢性缺氧组AE3mRNA的相对含量明显地低于对照组(P<005),慢性缺氧伴二氧化碳潴留及慢性呼吸性酸中毒组的AE3mRNA降低更为明显;(2)慢性缺氧伴二氧化碳潴留及慢性呼吸性酸中毒二组之间无显著差异(P>005),但都明显低于慢性缺氧组(P<001)。结论:提示AE3在呼吸衰竭及其导致的肺性脑病的发生中可能具有一定作用。
AIM:To study the expression of anion exchanger (AE 3) of brain in rats under chronic hypoxia and hypercapnia.METHODS:With a sensitive digoxigenin labelled AE 3 cDNA probe,dot blot was used to evaluated changes of AE 3 mRNA in rats models of chronic hypoxia,chronic hypoxia with hypercapnia and chronic acidosis established by pouring mixed gases into the chamber respectively as compared with control.RESULTS:It was found that the relative contents of AE 3 mRNA in rat brains in experimental group were significantly lower than that in control group( P <0 05),and the relative contents of AE 3 mRNA in rat brains of hypoxia with hypercapnia and chronic acidosis were also significantly lower than that of chronic hypoxia( P <0 01),but there was no significant difference between chronic hypoxia with hypercapnia group and chronic acidosis group( P >0 05).CONCLUSION:AE 3 may play an important role in the respiratory failure and pulmonary encephalophathy.
