摘要
为解决大叶榉组织培养中萌芽难、增殖系数低的难题,选取健康、饱满的顶芽为外植体,配合不同激素配比对顶芽进行诱导萌芽、增殖培养的研究。结果表明:不同培养基和激素浓度对顶芽萌芽和增殖培养均影响不同,其中最佳顶芽萌芽培养基是WPM+BA1.0 mg/L,萌芽率最高可达98.3%;最佳增殖培养基是WPM+4.0BA mg/L,最大增值系数为15。
By selcting healthy and plump tip buds of Zelkova schneideriana Hand.-Mazz.as explant,and matching different concentrations of hormone,the germination induction and multiplication culture of tip buds were studied.The experimental result indicates: the best media for shoot inducement was derived as WPM added with 1.0 mg/L of BA and the highest germination rate reached 98.3%;The bud cluster was multiplicated rapidly on the WPM medium with 4.0 mg/L of BA and the mean multiplication factor reached 15.
