脑通复方对血管性痴呆大鼠海马尼古丁受体的影响 被引量:4

Effect of Nao Tong Compound Recipe on the Expression of Nicotinic Acetylcholine Receptors of Hippocampal in Vascular Dementia Rats

摘要 目的观察脑通复方制剂对拟血管性痴呆(VaD)模型大鼠脑组织神经型尼古丁受体(Nicotinic acetylcholine recep-tors,nAChR)的影响,分析脑通复方制剂改善血管性痴呆学习记忆能力的作用机制。方法采用改良Pulsinelli四血管阻断(Pulsinelli's four-vessel occlusion,4-VO)法复制大鼠血管性痴呆模型,脑通复方制剂治疗后观察学习记忆能力,West-ern-blot法测定脑组织神经型尼古丁受体蛋白水平。结果脑通复方制剂可缩短实验大鼠逃避潜伏期,增加跨越平台次数,上调海马组织神经型尼古丁受体的表达。结论脑通复方制剂可通过上调nAChR表达,从而改善血管性痴呆学习记忆能力。 Objective To observe the influnce of Nao Tong Compound Recipe on the nicotinic acetylcholine receptors(nAChR) in rats with vascular dementia(VaD) and to explore the preventive effects of traditional Chinese medicine on spatial learning and memory deficits in rats with VaD.Methods VaD model was created with the reformed Pulsinelli 4-VO method.After establishing the model,the rats were treated with Nao Tong compound recipe.The ability of learning and memory of VaD rats were assessed by Morris Water Maze Test.The expression of Nicotinic acetylcholine receptors in hippocampus was detected by Western-blot method.Results The ability of learning and memory of VaD rats was improved and the expression of nAChR in hippocampus was increased after treatment with Nao Tong compound recipe.Conclusion By increasing the expression of nAChR and protecting neurons in hippocampus following ischemia,Nao Tong compound recipe may improve the ability of learning and memory of VaD rats.
