摘要
目的建立原百部碱在大鼠体内HPLC测定方法,研究原百部碱静脉注射后在大鼠体内的药动学特点。方法大鼠静脉注射原百部碱不同时间点眼眶取血,HPLC测定原百部碱的血药浓度,3P87软件处理数据。结果大鼠血清样品中原百部碱在0.10-12.60μg/ml范围内线性关系良好,大鼠静脉注射原百部碱后药时曲线符合双室模型,分布速率常数为2.69h^-1,消除速率常数为0.69 h^-1,AUC为3.82 mg.h.L^-1。结论所建立的血药浓度测定方法操作简便、准确灵敏、重现性好,可用于原百部碱在大鼠体内的药动学研究。原百部碱静脉注射后分布和消除均较快。
Objective To establish an HPLC method for the determination of protostemonine in rat plasma and to study pharmacokinetics parameters in rats after i.v.injection.Methods After taking protostemonine through i.v.injection,rat plasma samples were collected at different time points,and they were determined by HPLC and 3P87 to calculate data.Results The results showed that the linear range of calibration curve was within drug plasma concentration of 0.10-12.6 μg/ml,and the pharmacokinetics of protostemonine in rats were fitted to two-compartment model.Conclusion The method has good selectivity,sensitivity and reproducibility,which can be used for pharmacokinetic studies of protostemonine in rat plasma.Due to its fast absorption and elimination,it can be made into slow and controllable released preparations.
