复元胶囊对实验性大鼠骨关节炎软骨中TNF-α和VEGF的影响

Effects of FuYuan Capsule on Tumor Necrosis Factor-alpha and Vascular Endothelial Growth Factor in Cartiage Cells of Experimental Osteoporosis Rats

目的观察复元胶囊对实验性大鼠骨关节炎软骨中的TNF-α和VEGF的影响。方法 56只SD大鼠随机分为:正常对照组6只,生理盐水灌胃;模型对照组10只,造模后生理盐水灌胃;四环素组,复元胶囊低、中、高剂量组各10只,造模后分别用四环素、复元胶囊不同剂量灌胃。治疗10周后分别取各组胫骨平台关节软骨,采用HE染色进行Mankin’s评分,免疫组织化学染色法测定关节软骨中的TNF-α和VEGF表达的阳性细胞数。结果正常对照组HE染色Mankin’s评分与实验组比较,差异具有统计学意义(P<0.05)。各组关节软骨切片TNF-α和VEGF免疫组化染色分析,正常对照组TNF-α和VEGF与实验组比较,差异具有统计学意义(P<0.05)。结论复元胶囊可能通过调控TNF-α和VEGF抑制炎症和血管增生,从而达到延缓关节软骨退行性改变的作用。

Objective To observe the effects of FuYuan Capsule on tumor necrosis factor-alpha（TNF-α）and vascular endothelial growth factor（VEGF）in osteoarthritis（OA）.Methods Fifty-six Spragne Dawley（SD）rats,were randomly divided into five groups： normal group with 6 rats,model contrast group with 10 rats,tetracycline group with 10 rats,FuYuan capsule low,moderate and high does group with 10 rats.The normal group and model contrast group were fed with 0.9 N.S,tetracycline group were fed with tetracycline and others with FuYuan Capsule.After 10 weeks,the rats were sacrificed,and then rat tibiaarticular cartilage was removed.Sections of the articular cartilage were stained with hematoxylineosin.The cartilage histological changes were observed according to the method of Mankin.Immunohistochemical staining was performed to investigate the expression of TNF-α and VEGF.Articular cartilage were observed with microscope and image analysis method was used to measure the expression intensity of TNF-α and VEGF.Results There were significant differences in Mankin score among normal group and other groups（P〈0.05）.Immunohistochemical staining indicated that the expression intensity of TNF-α and VEGF among normal group and experimental groups were significantly different（P〈0.05）.Conclusion FuYuan capsule can protect the articular cartilage through suppressing the TNF-αand VEGF expression in chondrocyte,with suppressing inflammation and andangiogenesis.
