摘要
[目的]筛选山药茎尖脱毒苗分化培养基配方。[方法]选用MS为基础培养基,6-BA、2,4-D、NAA为激素处理因素和不同激素浓度处理水平的3因素3水平的试验设计,进行了山药茎尖脱毒苗分化培养基的筛选研究。[结果]MS+2.0mg/L6-BA+1.0mg/L2,4-D+3.0mg/LNAA为山药茎尖脱毒苗最佳分化培养基。[结论]该培养基能达到山药茎尖离体脱分化与分化之间的一种良好的平衡,是一种进行山药茎尖脱毒苗培养适宜的分化培养基。
[Objective]The research aimed to screen out the differentiation medium formula for the virus-free test-tube seeding from stem tips of Dioscorea opposita. [Method]Choosing MS as the basic medium,three-factor and three-level test was designed with 6-BA,2,4-D and NAA as treatment factors and different hormone levels as three levels. The screening of the differentiation medium for the stem tips of Dioscorea opposita was studied. [Result]MS +2. 0 mg/L 6-BA +1. 0 mg/L 2,4-D +3. 0 mg/L was the optimum differentiation medium for the virus-free seedling of Dioscorea opposita. [Conclusion]The medium could reach a favorable balance between dedifferentiation and differentiation of stem tips of Di-oscorea opposita. The medium was a kind of suitable differentiation medium for the virus-free seedlings of the stem tips of Dioscorea opposita.
