X线照射诱导人胃癌SGC-7901细胞生物特性改变以及YKL-40基因表达变化

Changes of biological characteristics and expression of YKL-40 gene in gastric cancer SGC-7901 cell induced by X-ray irradiation

摘要 目的观察X线照射后胃癌SGC-7901细胞形态改变、细胞周期分布情况以及是否可以诱导YKL-40基因的表达。方法将胃癌细胞株SGC-7901分为空白对照组(不照射)和实验组(2Gy/1f和4Gy/2f),实验组细胞经6-MVX线照射后取两组细胞在倒置显微镜下观察细胞形态学的改变,流式细胞术检测SGC-7901细胞周期的变化情况,半定量RT-PCR检测基因YKL-40mRNA的表达。结果实验组(2Gy/1f)细胞经X线照射后出现明显的形态学改变;流式细胞检测发现,与空白对照组细胞比较,2Gy、4Gy实验组细胞S期上调,G2/M期下调,组间比较差异有统计学意义(P<0.05)。RT-PCR检测胃癌细胞株SGC-7901不表达YKL-40或检测不到YKL-40mRNA,经X线照射后YKL-40表达。结论 X线照射可使人胃癌细胞发生形态改变及细胞增殖周期再分布,并且可诱导人胃癌细胞表达YKL-40。 Objective To observe the morphological changes,the division of cells cycle and expression of YKL-40 gene level in gastric cancer SGC-7901 cells after irradiation by X-ray.Methods SGC-7901 cells were devided into control group(no irradiation)and experimental group(2Gy/1fraction and 4Gy/2fraction).SGC-7901 cells in the two groups were taken.Morphological changes of the tumor cells by invert microscop,divisions of cell cycle by FCM and changes of YKL-40 mRNA of SGC-7901 cells by RT-PCR assay in experimental grop were observed compared to control group.Results Morphology of SGC-7901 cells in 2Gy/1 fraction group showed significant changes after irradiation.Cell S phase up and G2/M phase down in 2Gy/1 fraction group and 4Gy/2 fraction group,there was significant difference between the two groups(P〈0.05).Meanwhile,expression of YKL-40 mRNA could be induced by irridiation.Conclusion We find that human gastric cancer SGC-7901 cells changed in morphology and redistributed in cell cycle after irradiation of X-ray.Inaddition,it can increase the expression of YKL-40.
