摘要
利用聚乙二醇(PEG)与磷酸盐的合理配比组成的双水相体系能够使猪血超氧化物歧化酶(SOD)粗酶得到进一步纯化,研究结果表明,6%PEG-6000与24%磷酸盐(pH7,0)组成的双水相体系一次成相萃取,使猪血SOD粗酶的纯度提高3-5倍,活力回收70~95%,而且发现SOD在PEG/磷酸盐双水相体系中的分配行为有一定规律,使用双水相处理粗酶能有效、快速、大量使样品达到一定纯度.
Raw SOD enzyme extracted from pig blood can be further purified by means of aqueous two-phase system composed of polyethylene (PEG) and phosphate at rational proportioning, Results obtained have shown that an once-thruogh phasing extraction with an aqueous two-phase system of 6% PEG-6000 and 24% phosphate (pH=7, 0) is fast and effective enough to achieve purification of raw pig-blood SOD enzyme on a large scale up to an enrichment of 3 to 5 times, with an activity recovery between 70% to 95%,Some laws that govern the proportioning behavior between the PEG and phosphate in the aqueous two-phase system have been discovered too,
