摘要
研究了利用聚乙烯醇复合材料包埋简单节杆菌转化醋酸可的松脱氢的动力学过程.实验测得了醋酸可的松在Tris-HCl缓冲液中的饱和浓度和在该固定化细胞中的有效扩散系数.采用排除内、外扩散影响的方法建立了相应的动力学模型,计算值与实验值吻合较好.
The cells of arthrobacter simplex BY 2 3 51 having Δ 1 dehydrogenase activity were immobilized by entrapment with polyvinyl alcohol alginate complex gel.The kinetics of the transformation of cortisone acetate to prednisone acctate by the immobilized cells was investigated.The saturation concentration of cortisone acetate in Tris HCl buffer and the effective diffusion coefficient of cortisone acetate in the immobilized cells were measured respectively.The kinetic model was derived for removing the effects of internal and external diffusion.The parameters of the kinetic model were obtained in experimens.The good agreement between the theoretical prediction and experimental data indicates the reliability of the proposed model.
