五子衍宗丸对少精子症模型大鼠的实验研究 被引量:10

摘要 目的研究五子衍宗丸对腺嘌呤所致的少精子症模型大鼠的影响,探讨其改善男性少精子症的作用机理。方法 72只成年雄性大鼠随机分为6组:空白对照组,模型组,阳性药组及五子衍宗丸大、中、小剂量组。以腺嘌呤溶液(250 mg/kg)每日灌胃,连续4周,造成大鼠少精子症模型。同日各受试药组按剂量灌胃给药,连续4周。取附睾尾作精子计数、形态学观察及精浆中性α-糖苷酶测定;取血测肝、肾功能及生殖激素;肝、肾及生殖器官称重并计算脏器指数及病理形态学观察。结果与模型组比较,五子衍宗丸各剂量组精子数量和活动率显著升高,畸形率显著下降,并呈现量效关系;精浆中性α-糖苷酶显著升高;大、中剂量组肝、肾功能有显著改善;血清睾酮显著升高;附睾指数及前列腺+精囊指数显著增加。形态学观察显示:五子衍宗丸各剂量组肾脏病变明显改善。结论 五子衍宗丸对腺嘌呤诱发的少精子症模型大鼠有明显的改善作用。 Objective: To investigate the effects of "Wu Zi Yan Zong" recipe on rat model with oligozoospermia induced by adenine and its pharmacological mechanism of improving oligozoospermia. Methods. Seventy two male rats were randomly and equally assigned to 6 groups (12 for each group) .. blank-control, model, positive-control and 3 test groups at different doses of "Wu Zi Yan Zong". Adenine at dose of 250 mg/kg b.w. was given to rats by intragastric administration daily for 4 weeks except for blank-control group. Concurrently, positive control medicine or "Wu Zi Yan Zong" recipe were also given to rats daily by intragastric administration for 4 weeks. The rats were sacrificed after the last administration of the test drugs. Epididymal sperm concentration and morphology, seminal plasma neutral α-glucosidase, serum hepatic and kidney functions, and reproductive hormones were assayed. Liver, kidney and reproductive organ of rats were weighed and calculated for the ratio of organ to body weight. In addition, pathological morphology was also examined. Results. Epididymal sperm concentration and motility increased while sperm abnormality rate decreased in a dose-dependent manner in "Wu Zi Yan Zong" recipe test group. In addition, seminal plasma neutral α-glucosidase level increased in each "Wu Zi Yan Zong" recipe test group. Liver and kidney functions of rats at high or middle dose test group were significantly improved. Serum testosterone concentration and ratio of reproductive organ to body weight in all test groups were significantly increased. Morphological changes of kidney in all test groups were obviously improved.Conclusions: "Wu Zi Yan Zong" recipe treatment has an improved effect on rat model with oligozoospermia induced by adenine.
