鸡肾型传染性支气管炎病毒的分离及其N基因序列分析 被引量:2

摘要 通过病毒形态观察、血凝特性试验、病毒干扰实验、动物回归实验等分离鉴定了1株鸡肾型传染性支气管炎病毒,命名为LY07。利用RT-PCR技术对分离毒株的N基因进行了扩增,经克隆、序列测定和分析比较,结果表明,N基因全长为1230 bp,编码1条409个氨基酸组成的多肽;其核苷酸与H52、M41和BJ等毒株的同源性为84.4%~89.7%,氨基酸同源性为86.1%~91.0%;LY07株与LX4、BJ和GDS14株关系较近,与其余鸡传染性支气管炎病毒株的亲缘关系均较远,是1株新的肾型IBV毒株。 One strain of avian infectious bronchitis nephritis virus(IBV) was identified by morphological observation,inoculating virus into chicken embryos,vira1 interference test,HA test and returning pathogen to host.It was designated as LY07 strain.The N gene of the strain was amplified by RT-PCR,and it was proved to be the N gene of infectious bronchitis virus by cloning,sequencing,and analysis.The results showed that cDNA of N gene of LY07 was composed of 1230 nucleotides encoding a polypeptide of 409 amino acids.Nucleotide homogeneity of N gene of LY07 with N gene of other IBV strains H52,M41,BJ ranged from 84.4 %-89.7 %,while 86.1 %-91.0 % identity was found on amino acid level.Evolution analysis indicated that LY07 was nearly related to BJ,GDS14 and LX4 and far related to other IBV strains.These results suggested that IBV LY07 was a new variance of IBV.
