益智宁神口服液对抽动—秽语综合征模型大鼠神经生物学和行为学的影响 被引量:4

Biological and Neurobehavioral Study of Yizhi Ningshen Oral Liquid on Tourette's Syndrome Model Rats

摘要 【目的】观察益智宁神口服液对抽动—秽语综合征(TS)模型大鼠神经生物学和行为学的影响。【方法】选用SPF级SD大鼠60只,随机分为空白组、模型组、中药组(益智宁神口服液,剂量为2 mL/d)、西药组(氟哌啶醇,剂量为0.15 mg/d)。除空白组外,其他组均采用亚氨基二丙腈(剂量为150 mg.kg-1.d-1)腹腔注射复制TS大鼠模型,造模后第2天开始按设计剂量给药,观察各组大鼠运动行为及刻板运动评分,采用高效液相色谱法检测治疗14 d后各组大鼠脑组织多巴胺(DA)和去甲肾上腺素(NE)的水平。【结果】模型组运动行为、刻板运动积分以及脑组织NE水平均显著升高(P<0.01),DA水平显著降低(P<0.01);中、西药组治疗14 d后均可降低大鼠运动行为、刻板运动积分及脑组织NE水平,升高脑组织DA水平,与模型组比较差异均有显著性意义(P<0.05或P<0.01),提示中药组作用与西药组相仿。【结论】益智宁神口服液治疗TS的作用与其能调整中枢神经递质的水平有关。 Objective To investigate the biological and neurobehavioral effect of Yizhi Ningshen Oral Liquid(YNOL) on Tourette's syndrome(TS) model rats.Methods Sixty SD rats with specific-pathogen free were randomized into blank control group,model group,YNOL(2 mL/d) group,and haloperidol(0.15 mg/d) group.Except for the blank control group,the rats in other groups received intraperitoneal injection of iminodipropionitrile to induce TS,and then were treated with the corresponding medicine on the second day of the modeling.The scores of motor behavior and stereotyped movement were observed.After treatment for 14 days,the cerebral levels of dopamine(DA) and norepinephrine(NE) were detected by liquid chromatography in various groups.Results In the model group,the scores of motor behavior and stereotyped movement and cerebral NE level were increased(P〈0.01),and cerebral DA level was decreased(P〈0.01).YNOL and haloperidol decreased the scores and cerebral NE level,and increased cerebral DA level of the TS rats(P〈0.05 or P〈0.01 compared with those in the model group),the differences between YNOL group and haloperidol group being insignificant.Conclusion The therapeutic effect of YNOL for TS is related with the regulation of central neurotransmitters levels.
