摘要
目的通过检测乳腺癌病变中胰腺十二指肠同源盒-1(PDX-1)蛋白及mRNA的表达情况,分析其与乳腺癌分子亚型之间的关系。方法采用免疫组织化学EnVision法检测PDX-1、雌激素受体(ER)、孕激素受体(PR)、第二号人类表皮生长因子受体(Her-2)、细胞角蛋白5&6(CK5/6)和上皮生长因子受体(EGFR)蛋白的表达,实时定量RT-PCR法检测PDX-1mRNA的表达。结果 PDX-1蛋白和mRNA在乳腺癌管腔A型和管腔B型中的表达显著低于在Her-2过表达型、基底细胞样型和未分类型中的表达(P<0.05)。结论 PDX-1蛋白和mRNA在乳腺癌管腔A型和管腔B型中的表达显著降低,PDX-1有望成为乳腺癌基因分型的筛选新指标。
Objective To study the expression and the relationship of PDX-1 protein and mRNA in molecular subtypes of Breast Cancer.Methods Immunohistochemistry was performed to determine the expression of ER,PR,Her-2,EGFR,CK5/6 and PDX-1 protein,and Real-Time RT-PCR was performed to analysed PDX-1 mRNA in molecular subtypes of Breast Cancer.Results The expression of PDX-1 protein and PDX-1 mRNA in luminal subtype A and luminal subtype B was lower than those in Her-2 overexpression subtype,basal-cell like subtype and unclassified subtype（P〈0.05）.Conclusion It is possible that PDX-1 would be a new candidate gene in molecular subtypes of breast cancer.
