摘要
目的探讨硒结合蛋白(SBP)-1、Caspase-3、Ki-67在子宫内膜癌中的表达及其生物学意义,为子宫内膜癌的早期诊断及预后判定提供实验依据。方法用免疫组织化学Envision法检测SBP-1、Caspase-3、Ki-67在70例子宫内膜癌中的表达,并选取25例子宫内膜非典型增生和20名正常子宫内膜进行对照分析。结果在正常子宫内膜中SBP-1、Caspase-3、Ki-67阳性表达率分别为95%、100%、5%;在子宫内膜非典型增生中SBP-1、Caspase-3、Ki-67阳性表达率分别为80%、72%、60%;在子宫内膜癌中分别为60%、50%、74%。SBP-1、Caspase-3、Ki-67分别在正常子宫内膜、子宫内膜非典型增生和子宫内膜癌中的表达差异有统计学意义(P<0.05)。SBP-1在子宫内膜癌中的表达与年龄无关(P>0.05),与组织病理分级、临床分期和肌层浸润深度及淋巴转移有关(P<0.05)。Caspase-3在子宫内膜癌中的表达与年龄、临床分期和肌层浸润深度及淋巴转移无关(P>0.05),与组织病理分级有关(P<0.05)。Ki-67在子宫内膜癌中的表达与组织病理分级和临床分期、肌层浸润深度及淋巴转移有关(P<0.05),与年龄无关(P>0.05)。在子宫内膜癌中,SBP-1与Caspase-3的表达呈显著正相关(r=0.467,P<0.05),SBP-1与Ki-67的表达呈显著负相关(r=-0.414,P<0.05),Caspase-3与Ki-67的表达呈显著负相关(r=-0.523,P<0.05)。结论 SBP-1、Caspase-3、Ki-67可作为子宫内膜癌组织的标记物,其相关性有助于子宫内膜癌的早期诊断及预后判定。
Objective To investigate the expressions and biological significance of SBP-1, Caspase-3 and Ki-67 in endometrial cancer, so as to provide laboratory evidences for early diagnosis and prognosis estimation. Methods The expressions of SBP-1, Caspase-3 and Ki-67 in 70 cases of endometrial carcinoma were determined using im- munohistochemical envision methods, and compared with findings from 25 cases of atypical endometrial hyperplasia and 20 of normal endometrial tissues. Results The expressions of SBP-1, Caspase-3 and Ki-67 were positive in 95%, 100% and 5% of normal endometrium, 80%, 72% and 60% of atypical endometrial hyperplasia cases, 60%, 50% and 74% of endometrial carcinoma, respectively. These differences in expressions of SBP-1, Caspase-3 and Ki-67 in normal endometrium, atypical endometrial hyperplasia and endometrial cancer appeared significant (P0.05). The ex- pression of SBP-1 in endometrial cancer was related to histopathological grades, clinical stages, depth of myometrial invasion and lymph node metastasis (P0.05) but not to age (P0.05). The expression of Caspase-3 in endometrial car- cinoma was related to histopathological grades (P0.05) but not to clinical stages, depth of myometrial invasion, lymph node metastasis and age (P0.05). The expression of Ki-67 in endometrial carcinoma was related to histopathological grades, clinical stages, depth of myometrial invasion and lymph node metastasis (P0.05) but not related to age (P 0.05). In endometrial carcinoma, SBP-1 and Caspase-3 expressions were positively correlated (r=0.467, P0.05),SBP-1 and Ki-67 expressions negatively correlated (r=-0.414, P0.05), Caspase-3 and Ki-67 expressions negatively correlat- ed (r=-0.523, P0.05). Conclusion SBP-1, Caspase-3 and Ki-67 can be useful as biomarkers in endometrial can- cer, of which, correlations among these markers may contribute to early diagnosis and prognosis estimation.
