摘要
目的:了解错配修复基因hMSH6在胃癌中的表达特点。方法:用免疫组织化学SP法检测45例胃癌组织、36例癌旁黏膜和30例正常胃黏膜hMSH6蛋白的表达情况。结果:胃癌组、癌旁组与正常胃黏膜组hMSH6蛋白细胞核表达阳性率分别为24.44%(11/45)、13.89%(5/36)和53.33%(16/30);细胞质表达阳性率分别为66.67%(30/45)、58.33%(21/36)和96.67%(29/30);细胞核和细胞质表达阳性率胃癌组和癌旁组均显著低于正常胃黏膜组(P<0.05);而胃癌组与癌旁组hMSH6蛋白在细胞核及细胞质中表达差异均无显著性意义(P>0.05)。胃癌组织中,hMSH6蛋白在细胞核中表达与患者性别、年龄,肿瘤分化程度及分期、淋巴结转移均无显著相关性(P>0.05),但在细胞质的表达与肿瘤分化程度及分期正相关(P<0.05),而与患者年龄、性别、淋巴结转移无显著相关性(P>0.05)。结论:胃癌及癌旁组织均出现错配修复基因hMSH6表达下调,这可能将成为胃癌发生的预警信号。
Objective: To study the expression feature of mismatch repair gene hMSH6 in gastric carcinoma. Meth- ods:Immunohistoehemisty method was used to detect hMSH6 protein expression in 45 cases of gastric carcinoma,36 cases of carcinoma adjacent mucosa and 30 cases of non - carcinoma gastric mucosa. Results:The positive rate of hMSH6 in cell nucleus : carcinoma group 24.44% ( 11/45 ) , carcinoma adjacentmucosa group 13.89% ( 5/36 ) and non - carcinoma gastric mucosa group 53.33% ( 16/30 ) ; The positive rate in cytoplasm : carcinoma group 66.67% ( 30/ 45 ), carcinoma adjacent mucosa group 58.33% (21/36) and non - carcinoma gastric mucosa group 96.67% (29/ 30). The positive rate in both nucleu and cytoplasm were carcinoma and carcinoma adjacent mucosa groups significandy lower than that in non- carcinoma gastric mucosa group( P 〈0.05 ). While the difference was not significantly between these two groups( P 〉 0.05 ). In gastric carcinoma group, the positive rate of hMSH6 in nucleus was not related to sex and age of the patient, differentiated degree and lymphonode metastasis condition of the carcinoma( P 〉 0.05 ) ; and in cytoplasm the positive rate of hMSH6 was not related to sex and age of the patient, and lymphonode metastasis condition of the carcinoma but related to differentiated degree of carcinoma( P 〈 O. 05 ). Conclusion:The functional downexpression of mismatch repair gene hMSH6 may play some roles in the gastric carcinoma.
