胃癌组织中PTEN表达与血管生成和侵袭转移关系 被引量:10

Correlation of the quantitative expression of PTEN and angiogenesis with the invasion and metastasis of gastric cancers

摘要 目的:探讨胃癌组织中PTEN的表达及其与胃癌血管形成和侵袭转移的关系。方法:应用免疫组织化学SP法检测60例胃癌组织中的PTEN及血管内皮生长因子表达情况,应用图像分析技术分析PTEN在胃癌组织中的阳性表达率。按照Weidner方法计数胃癌组织的微血管密度值。结果:PTEN在胃癌组织中的表达及其免疫反应分值与分化程度、淋巴结转移及浸润深度密切相关(P<0.05)。在超过肌层、有淋巴结转移及低分化胃癌组织中的微血管密度明显高于浸润肌层及肌层以上、无淋巴结转移及高-中分化的胃癌组织(P<0.05)。在胃癌组织中微血管密度值在PTEN阴性组明显高于阳性组,且胃癌组织中PTEN的免疫反应分值与其微血管密度值呈负相关。结论:PTEN的丢失或低表达与胃癌侵袭转移密切相关,对促进胃癌组织中新生血管形成有一定作用,PTEN和微血管密度对判断胃癌患者预后及指导临床治疗有一定意义。 Objective To investigate the correlation of the expressions of PTEN and angiogenesis with the invasion and metastasis of gastric cancers.Methods The expressions of PTEN and vascular endothelial growth factor were evaluated in 60 patients with gastric cancers with immunohistochemical SP method.The technique of image analysis was used to analyze the expression rate of PTEN in gastric cancers.The microvascular density value was counted according to the method of Weidner.Results The expressions of PTEN and its immune response scores in gastric cancers were closely correlated to the invasion depth,lymph node metastasis and differentiation grade(P0.05).The microvascular density in invading serosa,lymph node metastasis and poor differentiation was significantly higher than that in invading mucosa,submucosa and muscularis,lymph node metastasis and moderate-high differentiation tissues in gastric cancer.PTEN was negatively correlated to microvascular density(P0.05),and the microvascular density value in the negative group was higher than that in the positive group.Conclusion The loss or the low expression of PTEN is closely correlated to the invasion and metastasis of gastric cancers and promotes their angiogenesis.PTEN and microvascular density can be used to judge the prognosis of gastric cancer and guide the therapy treatment.
