降糖益肾方对高脂饲养MKR鼠糖代谢、肾功能的影响

Effects of Jiangtang Yishen decoction on glucose metabolism and renal function in MKR mice fed with high fat food

目的观察降糖益肾方干预MKR转基因2型糖尿病小鼠的血糖代谢、尿微量白蛋白(UmAlb)、β2微球蛋白(β2-MG)及血清尿素氮(BUN)、肌酐(Cr)的变化。方法取MKR小鼠(12周龄)40只经鉴定后,随机分为MKR鼠组、MKR鼠高脂组、降糖益肾方组、糖适平合用贝那普利对照组(简称西药组),每组10只,另设10只同周龄C57小鼠为空白对照组。MKR鼠组、空白组予以基础饲料喂饲,MKR鼠高脂组、降糖益肾方组、西药组予以高脂饲料喂饲。降糖益肾方组以62.4 g/kg浓度,西药组给以糖适平9.09 mg/kg、贝那普利3.04 mg/kg浓度灌胃,连续给药30 d,MKR鼠组、MKR鼠高脂组和空白组以等容量的蒸馏水灌胃。采用电化学法检测血糖,生化法检测血清BUN及Cr,双抗体夹心ELISA法分别检测血胰岛素、尿UmAlb及β2-MG含量。结果 MKR高脂组小鼠具有高空腹血糖,显著增高的胰岛素、血清BUN、Cr及尿UmAlb、β2-MG水平,与MKR鼠组比较,差异具有统计学意义(P<0.01)。给药30 d后,与MKR高脂组比较,降糖益肾方组小鼠的血糖水平、血清BUN、Cr及尿UmAlb、β2-MG的含量均显著下降,高胰岛素血症得到改善,差异具有统计学意义(P<0.05,P<0.01);且降糖益肾方优于或与西药组作用相当。结论降糖益肾方能显著改善高脂喂饲的MKR鼠空腹血糖,改善高胰岛素血症,降低其血清BUN、Cr及尿UmAlb、β2-MG的含量,改善高脂喂饲的MKR鼠的早期肾损伤,从而延缓高脂喂饲的MKR鼠肾脏病变的进一步发展。

Objective To observe the effect of Jiangtang Yishen decoction（JYD） on the levels of blood glucose,urine UmAlb,urine β2-MG,serum BUN and serum Cr in MKR mice with diabetes type2.Methods All 40 mice were randomly divided equally into four groups as fllows：ordinary feed group（OFG）,high fat feed model group（MG）,JYD group and posi-tive control group（PCG）,and another 10 C57 mice was the control group.The mice in JYD and PCG were fed with corresponding drugs for 30 days,in OFG,MG and PCG with the same dose of distilled water.The levels of serum insulin,urine UmAlb and β2-MG were measured with double-antibody sandwich ELISA,the serum BUN and Cr with biochemical,and the blood glucose with electrochemical detection method.Results Compared with MG group,the contentof blood glucose,serum insulin,urine UmAlb,β2-MG,serum BUN and Cr in JYD group signifi-cantly decreased（P0.01）;after 30 days,the content of blood glucose,serum insulin,urine UmAlb,β2-MG,serum BUN and Cr in JYD group were decreased obviously and hyperinsulinemia was improved（P 0.05,P 0.01）.Conclusion JYD can significantly improve the fasting blood glucose and hyperinsulinemia,delay development of renal functions in MKR mice fed with high fat food,
