摘要
为了研究纤维素酶的酶学性质,以产纤维素酶的绿色木霉为出发菌株,采用微波诱变处理,对其发酵生产的纤维素酶酶学性质进行研究。结果表明,绿色木霉经微波诱变与原有菌种相比产酶活力提高30%以上;该酶分别在37℃和pH 5.0时,酶活性最高;在羧甲基纤维素底物保护下,酶的热稳定性大幅度提高;金属离子Ba2+、K+对酶有激活作用,Cu2+、Ca2+、Fe2+、Mn2+、Zn2+、Fe3+对酶有抑制作用。
The cellulase-producing Trichoderma viride stains were treated with microwave to study zymology property of cellulase.The results showed that the activity of cellulase-producing Trichoderma viride stain induced by microwave was 30% higher than that of origin Trichoderma viride stain,the cellulase activity was the highest at 37℃ and pH 5.0,the thermal stability of cellulase was greatly increased under protection of CMC,Ba2+ and K+ had activation effect on cellulase activity but Cu2+,Ca2+,Fe2+,Mn2+,Zn2+ and Fe3+ had inhibition effect on cellulase activity.
