摘要 目的比较紫外分光光度计与酶标仪测定盐酸川芎嗪含量的优缺点。方法分别用两种方法测定盐酸川芎嗪的含量。结果两种方法对盐酸川芎嗪含量的测定结果没有显著差异。酶标仪测定速度快,试剂用量少,但300nm以下96孔板的紫外吸收和加样量差异会增加测量误差。紫外标准曲线的斜率明显高于酶标仪,说明紫外的吸光度值对浓度的敏感程度高于酶标仪。结论两种方法在盐酸川芎嗪含量测定中各有优缺点,应根据实验对象选用合适的方法 ,有利于实验顺利开展。 Objective To compare the advantage and disadvantage of UV spectrophotometry and microplate reader for the assaying of Tetramethylpyrazine hydrochloride. Methods The content of Tetrarnethylpyrazine hydrochloride was detected by UV spectrophotometry and microplate reader,respectively. Results There was no significant difference of the measurement results between the two methods. Microplate reader was faster and needed fewer samples than UV spectrophotometry, but the UV absorption of 96 - well plates under 300 nm and the difference of sample amount would increase measurement error. The slope rate of standard curve of UV spectrophotometry was higher than that of microplate reader,which demonstrated that UV Spectrophotometry had higher sensitive to the concentration of samples than microplate reader. Conclusion UV spectrophotometry and microplate reader have respective advantages and disadvantages, Thus,we should select an appropriate method according to the samples and lab conditions.
