摘要
目的探讨第10号染色体缺失的磷酸酶和张力蛋白同源物基因(PTEN)、K-ras蛋白表达与卵巢子宫内膜异位症(内异症)恶变的关系。方法应用免疫组化方法检测21例内异症相关卵巢癌、10例不典型内异症及23例内异症病人PTEN、K-ras蛋白表达,同时结合内异症相关卵巢癌临床病理学参数,分析其在内异症恶变过程中作用。结果内异症相关卵巢癌组PTEN蛋白表达阳性率低于内异症组,差异有显著性(P=0.011);内异症相关卵巢癌组K-ras蛋白阳性表达率高于不典型内异症组及内异症组,差异均有显著性(P=0.002、0.023)。PTEN、K-ras蛋白表达与内异症相关卵巢癌的分化程度、临床分期具有相关性(P=0.007~0.047),而与年龄、病理类型无相关性(P〉0.05)。结论 PTEN蛋白表达缺失可能是卵巢内异症恶变过程中的早期事件,而K-ras蛋白的过表达可能与卵巢内异症的恶变有关,PTEN、K-ras的异常表达可能均参与了卵巢内异症的恶变过程。
Objective To investigate the association of abnormal expressions of PTEN and K-ras in the process of malignant transformation of endometriosis. Methods The protein expressions of PTEN and K-ras were detected with immunohistochemistry in 21 cases of endometriosis-associated ovarian cancers(EAOCs),10 of atypical endometriosis(aEMs) and 23 of endometriosis.The role of PTEN and K-ras in malignant transformation of endometriosis was analyzed in combination with the pathological and clinical data of EAOCs. Results The positive rate of PTEN expression in EAOCs was significantly lower than that in endometriosis(P=0.011).The positive rate of K-ras expression in EAOCs was significantly higher than that in aEMs and endometriosis(P=0.002,0.023).The expressions of PTEN and K-ras were significantly associated with the differentiation and clinical stage of EAOCs(P=0.007-0.047),but was not related to patient's age and pathology type(P0.05). Conclusion Reduction of PTEN protein expression might be an early event in malignant transformation of endometriosis,while over-expression of K-ras might be associated with the process of malignant transformation of endometriosis.Both PTEN and K-ras might be involved in malignant transformation of endometriosis.
