摘要
目的探讨白细胞介素-6(IL-6)对小鼠NIH3T3成纤维细胞中基质金属蛋白酶-3(MMP-3)、金属蛋白酶组织抑制剂1(TIMP-1)表达的影响。方法应用不同浓度(0、5、10、15、20 mg/L)的IL-6刺激NIH3T3成纤维细胞,共同培育12 h收集细胞。应用Western blot和RT-PCR方法分别检测细胞内MMP-3和TIMP-1的表达。结果 MMP-3表达随着IL-6浓度的升高而上升,TIMP-1表达随着IL-6浓度的升高而下降。结论 IL-6能影响MMP-3和TIMP-1的表达平衡,可能是影响硬膜外瘢痕形成的机制之一。
Objective To study the affect of IL-6 on the expressions of MMP-3 and TIMP-1 in NIH3T3 fibroblasts in mice. Methods Different concentrations(0,5,10,15 and 20 mg/L) of IL-6 were applied to stimulate NIH3T3 fibroblasts and culture for 12 h and collect the cells.Western bolt and RT-PCR were employed to detect the expressions of MMP-3 and TIMP-1 within the cells. Results The expression of MMP-3 increased with the rising concentration of IL-6,and TIMP-1 declined.Conclusion IL-6 can affect the balance between the expressions of MMP-3 and TIMP-1,which is,probably,one of the mechanisms of epidural scar formation.
