白花除虫菊的种质保存研究

摘要 本实验目的是获得对植物损伤尽可能小,保存时间尽可能长,保存后的白花除虫菊试管苗可以快速恢复并大量繁殖的种质保存条件。本实验通过改变白花除虫菊培养温度、添加植物生长抑制剂,调整培养基渗透压、营养水平等方法,对白花除虫菊进行种质保存实验。结果显示除虫菊在4℃下保存时间为6个月;在MS+脱落酸(ABA)1.0 mg/L+多效唑(PP333)1.0 mg/L+矮壮素(CCC)10.0 mg/L的培养基中,保存时间超过了5个月;降低蔗糖的添加量,可有效延缓白花除虫菊试管苗的生长速度。实验结果表明,常低温保存、添加植物生长抑制剂ABA、降低培养基营养水平等方法都可以有效延缓除虫菊的生长速度。 The experiments of germplasnl conservation of Chrysanthemum cinerariifolium were carried on to obtain a conservation condition. The conditions should have a low toxicity to the test tube plantlets and a conservation time as long as possible. The factors of these experiments need to be considered including culturing temperature, plant growth rctardant, the osmotic pressure and components of culture medium etc. The results indicated thai the conservation time of test tube plantlets of Chrysanthemum cinerariifalium was 6 months at 4 ℃. The test-tube plantlets were sensitive to abscisic acid(ABA), and the conservation time was more than ,5 months in the culture medium(MS+ABA 1.0 mg/L3 PP333 1.0 mg/L+ CCC 10.0 mg/L). The growth rate of test tube lolantlets could be put off effectively by reducing the addition of sucrose. The growth rate of test tube plantlets could be put off effectively by culturing at the low temperature, adding the plant growth retardant ABA, reducing the addition of sucrose etc.
