摘要
采用茎尖分生组织培养技术,获得了百合(Liliumbrownii F.E.Brown var.Viridulum Baker)的无病毒试管苗。通过基本培养基和激素配比试验,筛选出最佳的培养基组成,进行脱病毒苗的快速繁殖,并对试管苗进行生理生化指标及生物量的测试。结果表明:诱导丛生芽的最适培养基为:MS+BA(6-苄氨基嘌呤)1.0 mg/L+IAA(吲哚乙酸)0.2 mg/L+PP333(多效唑)4.0 mg/L+KT(激动素)1.0 mg/L,技术上达到了快速繁殖规模生产的要求。诱导试管苗最适生根的最适培养基为1/2 MS+IBA(鹅膏蕈氨酸)0.5 mg/L。通过电子显微镜的反复检测,筛选出10个彻底脱病毒的品种,这些品种可以为今后的无病毒优质种苗提供原种苗。发现检测的脱病毒百合试管苗的蛋白含量升高,SOD酶活升高,APX酶活升高,鲜重升高,预示脱病毒材料可能具有较好的生长势,对提高百合的产量有利。
Virus-free materials of Lilium spp. were obtained trom meristem culture in vitro. A series of optimization experiments for cultural medium composition and concentration of phytohormones were conducted with a view to accelerate the propagation of virus free materials. The best condition of chromosome determination was established. The obtained results indicated that the most suitable rapid propagation media of Lililum spp. was MS+ 6-BA 1.0 mg/L+ IAA0.2 mg/L+ KT1.0 mg/L+ PP333 4.0 mg/L. The optimized medium for rooting was 1/2 MS+ IBA 0.5 mg/L. These results could meet the technical requiremcnts of rapid propagation in large scale production. Virus free materials were observed repeatedly under electron microscope, 10 virus-free samples were screened out which could be used to the fast propagation for production. Some physiological data were tested among the virus-free lines and control lines. There was difference between the lines.
