摘要
目的:优化人心房组织膜蛋白提取方法。方法:分步离心法提取心肌膜蛋白,SK2膜蛋白用western-blot检测。结果:BCA检测出蛋白浓度,western-blot检测出蛋白条带。结论:此方法能够提取人心房组织的膜蛋白SK2,该方法简单,稳定,可靠,提取的膜蛋白质量好,浓度高,并能进行下一步的western-blot的实验,以便将来检测SK2膜蛋白在窦性和房颤患者心房组织之间表达的差异。
Objective: To optimize the method for extracting and solubilizing membrane proteins from human atrial tissue. Methods: Low-speed centrifugation followed by ultracentrifugation was applied in extracting membrane proteins. SK2 membrane protein was assessed by western blot. Results: Protein was measured by BCA with an average concentration of 1.442mg/ml. Protein bands were detected by western-blot successfully. Conclusion: Compared with other extraction method, this method is simple, steady and reproducible. Moreover, the quality of membrane protein is good and the concentration is high. Our research suggests that the membrane proteins extracted by the above-mentioned method are appropriate for further studying the expression difference of SK2 between the patients with sinus rhythm and the patients with atrial fibrillation.
