摘要
目的:研究顺铂(DDP)对昆明小鼠体内眶内组织巨噬细胞的作用。方法:用Griess法测定昆明小鼠体内眶内组织巨噬细胞一氧化氮(NO)的生成量,测定酸性磷酸酶(ACP)的活性,用免疫组织化学法测定巨噬细胞内乳酸脱氢酶(LDH)的表达。结果:DDP在120~180mg/ml浓度范围内可显著促进正常巨噬细胞NO的生成,在200~300mg/ml浓度范围内可抑制脂多糖(LPS)激活的巨噬细胞的NO生成。结论:DDP对昆明小鼠眶内组织巨噬细胞NO的生成具有激活和抑制双重作用。
Objective:To investigate the effects of Cisplatin on the peritoneal macrophages of Kunming mice.Methods:Nitric oxide produced by Kunming mice peritoneal macrophage was detected by Griess reagent,acid phosphatase in the macrophages was detected and analyzed.The activity of lactate dehydrogenase was determined by immunohistochemical staining.Results:NO was produced when macrophages were exposed to Cisplatin ranging from 120 to 180 mg/ml.Polysaccharide of Cisplatin ranging from 200 to 300 mg/ml showed a significantly inhibitory effect on LPS-induced NO production by macrophages.Conclusion:Polysaccharide of Cisplatin has a biphasic regulatory effect on NO production by Kunming mice peritoneal macrophages.
