摘要
目的探讨低频电刺激(LFS)丘脑底核对癫大鼠海马谷氨酸(Glu)及γ-氨基丁酸(GABA)表达的影响。方法用30只大鼠制备杏仁核电刺激点燃模型,同时在丘脑底核埋置刺激电极;制模成功的大鼠随机分为LFS组及对照组;两组大鼠每日点燃1次,共10d;LFS组大鼠每次点燃前予以LFS丘脑底核。观察10d后两组大鼠癫发作程度和持续时间,应用免疫组化法测定大鼠海马Glu、GABA阳性细胞数及灰度值。结果与对照组比较,LFS组癫发作的Racine评分明显降低,癫发作持续时间明显缩短;海马GABA阳性细胞数明显增多、灰度值降低;Glu阳性细胞数明显减少,灰度值明显增加(均P<0.05)。结论 LFS丘脑底核能有效抑制大鼠杏仁核点燃发作,其作用机制可能是改变了脑内Glu、GABA的表达。
Objective To investigate the influence of low frequency electrical stimulated(LFS) on sub-thalamic nucleus on expression of glutamate(Glu) and gamma aminobutyric acid (GABA) in the hippocampus in epilepsy rats.Methods The amygdale electrical stimulation kindling models were made; meanwhile,the exciting electrode were embedded in discus lentiformis. The succeed models were divided into the LFS group and control group. The rats in the 2 groups were kindled completely once a day and last for 10 days. The rats in LSF group were received discus lentiformis LFS before kindling. After 10 days,the grade and duration of seizure were observed. The positive cell number and gray scale of Glu and GABA in the hippocampus were detected by immunohistochemical method.Results Compared with the control group,the score of Racine scale in LFS group was obviously decreased and seizure duration was significantly cut down. The number of GABA-positive cell was significantly increased but the gray scale was decreased; meanwhile,the number of glutamate-positive cell was decreased and gray scale was significantly increased (all P0.05).Conclusion LFS can be significantly suppress the seizures of amygdala kindling rats,the mechanisms may be involve the change of expression of Glu and GABA in brain.
