摘要
目的研究不同条件的MEK抑制剂U0126对食管癌Eca109细胞生长的影响。方法 Western-blot检测不同浓度的抑制剂U0126对ERK1/2表达的影响;MTT检测抑制剂U0126在不同的作用条件下,对食管癌Eca109细胞生长的影响并筛选出最佳的作用条件;流式细胞术检测U0126作用后细胞的凋亡情况。结果 Western-blot检测不同浓度的U0126对ERK1/2表达抑制作用,提示最佳抑制浓度为50μmol。MTT检测结果为不含胎牛血清(FCS)的RPMI1640配制U0126,作用1小时对食管癌细胞的生长抑制最佳。流式细胞术检测显示细胞凋亡率为6.5%。结论不同作用条件下的抑制剂U0126,对食管癌细胞生长的影响不同。
Objective To investigate effect of MEK inhibitor U0126 on cell growth in esophageal cancer 109 cell line under different conditions.Methods The expression of ERK1/2 with U0126 different concentrations were analyzed by Western-blot,the cell growth was analyzed by MTT after treated with U0126 under different conditions.The apoptosis of cell was detected by flow cytometry.Results The most effective inhibition concentration of U0126 was 50 μmol that was resolved with RPMI1640 without the content of fetal calf serium(FCS).The growth inhibition of esophageal cancer cell was analyzed by MTT after treated with U0126 1 hour when was the best time for growth suppression.The rate of cell apoptosis was about 6.5%.Conclusions Different conditions of MEK inhibitor U0126 have different effects on cell growth in esophageal cancer 109 cell line.
