大花朱顶红鳞茎不定芽的诱导 被引量:12

摘要 以大花朱顶红‘Red Lion’鳞茎作外植体,研究了最佳外植体消毒方式、最适不定芽诱导的培养条件和抑制外植体褐变现象的理想处理方法,以期为朱顶红工厂化生产提供理论依据。结果表明:最佳外植体消毒方法为0.1%升汞浸泡8min,污染率仅为5.0%,外植体正常生长;最适不定芽诱导培养基为MS+6-BA2.0mg/L+NAA1.0mg/L+3%蔗糖+Vc0.2g/L,可直接诱导成完整植株;Vc对外植体褐变的抑制效果好于活性炭(AC);将培养60天左右的试管苗移栽于草炭、珍珠岩和蛭石体积比为1:1:1的混合栽培基质中,成活率达100%。 The bulblets in vitro of Hippeastrum vittatum were used for explants to develop the tissue culture on MS,the research aimed to find the optimum culture conditions on adventitious bud induction.The results showed that to sterilize the explants by 0.l% density of mecruric chloridein for 8 min was the best with the contamination rate 4% only and the explants was in normal state,the optimum culture medium was MS+6-BA 2.0 mg/L+NAA mg/L+sucrose 5%+Vc 0.2 g/L,in which the complete plant was induced directly.Vc has better effect on inhibiting browning than AC.The survival rate was up to 100% when the seedlings,which were cultured 60 days,were transplanted to the matrix of peat,vermiculite,perlite with volume ratio 1:1:1.
