摘要
目的观察新型可降解镁锌合金(Mg-6wt.%Zn)对前成骨细胞MC3T3-E1细胞整合素α5表达的影响。方法将MC3T3-E1细胞接种于镁锌合金和左旋聚乳酸上,共培养1、3、6、9、12和15d后,收集细胞,利用实时荧光定量PCR(real-time PCR)方法检测成骨细胞整合素亚基α5mRNA的表达水平。结果镁锌合金表面Itgα5mRNA表达水平随着时间延长而增高(P<0.05),除第1天外,PLLA表面Itgα5 mRNA表达水平均低于镁锌表面(P<0.05)。结论与左旋聚乳酸相比,镁锌合金能够提高成骨细胞Itgα5 mRNA表达水平,有利于促进成骨细胞与材料表面的粘附。
Objective To observe the effect of Mg-Zn alloy on the integrin α5 expression of preosteoblasts MC3T3-E1.Methods The integrin α5 mRNA expression of MC3T3-E1 cultured with Mg-Zn and PLLA was estimated by real-time fluorescent quantitative polymerase chain reaction(real-time PCR) after 1,3,6,9,12 and 15 days.Results The mRNA expression of Itgα5 on Mg-Zn increased with time(P0.05),and the mRNA expression of Itgα5 on PLLA was totally lower than that on Mg-Zn except the first day(P0.05).Conclusion Compared with PLLA,Mg-Zn enhanced the mRNA expression of itgα5 of osteoblasts,which benefited to the attachment of osteoblasts with the materials surface.
