摘要
目的以不同剂量重组四价Aβ1-15腺病毒疫苗(Ad-4×Aβ15)接种于BALB/c鼠,寻求Ad-4×Aβ15疫苗的最佳免疫剂量并观察免疫效果,为疫苗的临床应用奠定基础。方法不同剂量Ad-4×Aβ15鼻黏膜途径接种BALB/c鼠,间接ELISA法检测血清中抗体的滴度的变化情况。Western blot法及Tg2576鼠脑切片免疫组化检测血清中抗体特异性,并对疫苗安全性进行评价。结果109pfu组和108pfu组两者平均抗体滴度差异无统计学意义(P>0.05)。109pfu组及108pfu组与107pfu组的抗体滴度相比较,差异有统计学意义(P<0.05)。BALB/c小鼠血清中产生的抗体能与Aβ42及Tg2576转基因鼠脑组织内老年斑特异性结合。疫苗接种后未导致小鼠脑、肝、肾病理损害。结论从首次免疫开始,109pfu组和108pfu组两者抗体平均滴度差异无统计学意义(P>0.05),但均与107pfu组差异有统计学意义(P<0.05)。从安全性上,108pfu为小鼠最合适免疫剂量。
Objective To inoculate BALB/c with the vaccine, exploring the best dose of immunity and safety of it in adult BALB/c and providing scientific bases for the clinical application of it.Methods Inoculate BALB/c mice with different dose of Ad-4×Aβ15 by nasal way.The change of the antibodies in the blood serum was detected by using the sandwich elisa , the specialty of the antibody could be identified by the western blot and the immunhistiochemistry , it's safety.Results There was no statistics difference between the 10^9pfu group and 10^8pfu group but there was statistics difference between 10^7pfu and other two groups.The specialty of the antibody could be identified by the Western blotting and the immuno-histiochemistry.The Aβ plaques were stained.There were no pathological changes in microstructure of liver , kidney and brain.Conclusions There is no statistics difference between the 10^9 pfu group and 10^8 pfu group at the antibody titre but there is statistics difference between 10^7pfu and other two groups.Think about the safety, 10^8 pfu is a better dose.
