摘要
采用新型超宽频带横电磁传输室BTEMCel,在低强度脉冲电场(WEMPF,电压187V,场强20V/cm,频率300Hz,脉宽50ns或100ns)作用下,鸡红细胞膜上出现直径0.1~0.9μm大小不等的孔洞,被穿孔的细胞约占所观察细胞的1%,细胞穿孔率与照射时间无明显相关.另外,还观察到有1%的细胞虽未穿孔但有小泡从膜上凸出或出现凹陷;台盼蓝能进入人红细胞,红细胞体积都明显变大;WEMPF能诱导枯草杆菌对卡那霉素产生抗性并稳定遗传,能引起细胞融合的频率约为1%.
Four experiments have been sep up to study whether the WEMPF elicits effects of electroporation,electrotrasfection and eletrofusion or not.1.Fixatation by glutaraldehyde before finishing exposure and then directly observ the surface of the chicken erythocyte through SEM. 2. By tracing Trypan blue as an indicator to observe whether small molecular enter the human erythocyte.3.Bacilluse Subtibis eletrotrasfection.4.Chicken erythocyte eletrofusion.It’s found that:1. After exposure,proes(0.1~0.9μm,esp.1.8μm),bubbles or concave are observed in the membrane surface,which is 1% and have no apparent relation to time of exposure.2. Trypan blue enter human erythrocyte while all of them become large obviously.3. WEMPF fail to result in eletrotrasfection in this condition but unexpectedly induce Kan resistance which has genetic stability.4. WEMPF can induce eletrofusion,however,probability is low,about 1%.
