幼龄大鼠哮喘气道重塑模型的构建

摘要 目的:幼龄大鼠哮喘气道重塑模型构建。方法:40只幼龄雄性SD大鼠随机分成正常对照组、哮喘模型组,每组20只。采用低剂量0VA,并用新鲜配制的氢氧化铝凝胶、百日咳杆菌菌苗作为免疫佐剂,经尾静脉注射和腹腔注射,2次致敏,并尾静脉注射维持致敏。结果:幼龄大鼠明显的呼吸困难,病理结果显示气道壁明显增厚,管腔变窄,气道黏膜皱襞增多,上皮细胞脱落,气道平滑肌层和网状基底膜层增厚,黏膜下及管壁大量嗜酸细胞、淋巴细胞浸润。讨论:实验证明改良后致敏效果良好,采用雾化吸入0VA进行连续激发,尾静脉注射维持致敏,成功地构建了幼龄大鼠哮喘气道重塑模型。 Objective:The young age big mouse asthma gas channel remoulds the model construction. Methods: 40 young age male SD big mouse divides into the normal control group,'asthma model group stochastically, each group of 20. Uses low dose OVA,and with the new configuration aluminum hydroxide gelatin,the bacillus pertussis vaccine took the immunity adjuvant,after the tail intravenous injection and the abdominal cavity injection, 2 times sends sensitively,and the tail intravenous injection maintenance sends sensitively. Results: The young age big mouse obvious breath difficulty, the pathology result demonstration gas channel wall obvious accumulation, the lumen stricts, the gas channel mucous membrane wrinkle increases, the epithelial ceil falls off, gas channel smooth muscle level and netted basis film accumulation,under mucous membrane and pipe wall massive oxyphils,lymphocyte infiltration. Discussion:The experiment after proved the improvement sends the sensitive effect to be good, uses atomization inspiration OVA to carry on stimulates continuously, the tail intravenous injection maintenance sends sensitively, successfully constructed the young age big mouse asthma gas channel to remould the model.
