摘要
目的研究肠道病毒71型(EV71)手足口病患者外周血淋巴细胞对不同EV71抗原刺激的增殖反应。方法将38例研究对象分为手足口病EV71阳性重症组8例、轻症组10例、EV71阴性组10例、健康对照组10例。外周血单核细胞(PBMC)与EV71合成多肽SP1、SP2、SP3、灭活EV71和植物血凝素(PHA)分别共同孵育,加入胸腺嘧啶核苷(3H-TdR),然后收集细胞于液闪仪测定每分钟脉冲数(cpm)。结果对不同EV71抗原的淋巴细胞增殖反应发现,以灭活病毒EV71免疫原性最强,SP2次之。EV71轻症和重症患者淋巴细胞增殖反应比较差异无统计学意义(P>0.05),EV71阴性手足口病患者及健康对照组对EV71抗原未能显示有效的淋巴细胞增殖反应。结论灭活EV71免疫原性最强,感染EV71的患者淋巴细胞对EV71抗原刺激呈特异性增殖反应。
Objective To investigate proliferation response of peripheral blood mononuclear cells ( PBMC ) of patients with HFMD infected with enterovirus 71 ( EV71 ) different EV71 antigens stimulation. Methods Peripheral blood mononudear cells ( PBMC ) were co - cultured with EV71 antigens peptide SP1, SP2, SP3, inactivated EV71 and phytohemagglutinin ( PHA ), respectively, thymidine ( 3 H - TdR ) was added and counts per minute ( epm ) were measured. Results Based on cpm, inactivate EVT1 is the strongest immunogenie antigen, followed by SP2. Lymphocyte activation between mild and severe EV71 have no significant differences( P 〉0.05 ) , and EV71 antigen stimulation had no effect on proliferation response in patients with HFMD uninfected with EV71 and the control group. Conclusion Inactivate EV71 is the most immunogenie antigen. Patients infected with EV71 have more specific lymphocyte proliferation response compared with controls and patients with EV71 infection.
