摘要
利用19个微卫星基因座位对52只圈养猎豹基因组DNA进行了扩增。19个基因座的等位基因数为2~11个,共检测到131个等位基因。基因杂合度为0.500 0~0.904 0,平均为0.741 6;多态信息含量为0.375 0~0.895 8,平均为0.699 7;有效等位基因数为2.000 0~10.440 2,平均为4.724 6。表明19个微卫星基因座在研究样本中均为中高度多态性基因座,显示出群体丰富的遗传多样性。运用软件SPSS12.0对52只猎豹个体间的亲缘关系进行聚类。结果显示,个体间亲缘关系清楚可见,在距离系数为4.520水平上可聚为12类。个体间的遗传距离聚类结果与系谱资料记录的结果基本一致。
Genome DNA of 52 captive Acinonyx jubatus were amplified by nineteen microsatellite loci.The amplification results showed 131 alleles were found and the number of alleles at each locus ranged from 2 to 11.The gene heterozygosity(He) ranged from 0.500 0 to 0.904 0 and loci FCA094 and FCA077 had the highest and lowest value,respectively.The polymorphism information content(PIC) ranged from 0.375 0 to 0.895 8 and loci FCA094 and FCA077 had the highest and lowest value,respectively.The effective number of allele(Ne) ranged from 2.000 0 to 10.440 2 and loci FCA094 and FCA077 had the highest and lowest value,respectively.These results showed that the 19 microsatellite loci in these materials were high or medium polymorphic,indicating high genetic diversities.The cluster analysis using SPSS12.0 software revealed that 52 members of Acinonyx jubatus were divided into twelve categories at 4.520 level.Most of the relationship among the individuals were clear.
