摘要
目的寻找适合原子力显微镜(atomic force microscopy,AFM)观察甲状腺活体组织细胞标本的最佳制备方法。方法选用压片组织细胞法、细胞印片法和酶消化离心涂片法制备适合于AFM观察的活体甲状腺组织细胞样品,对比观察三种方法制备的细胞样品在光学显微镜和AFM下的细胞分散度和图像清晰度,从而找出适合AFM观察甲状腺活体组织细胞样品的最佳制备方法。结果细胞印片法制备的甲状腺细胞样品在AFM对其进行扫描时,很容易找到单个细胞,杂质较少、干扰较小,且细胞表面形貌成像清晰,其他两种方法制备效果不理想。结论细胞印片法是最适合AFM观察甲状腺活体组织细胞样品的制备方法;利用AFM扫描活体甲状腺组织细胞的纳米级结构是确实可行的,为进一步利用AFM研究正常甲状腺细胞和肿瘤性甲状腺细胞提供了可能。
Objective To seek the best method for preparation of thyroid biopsy specimens under the atomic force microscopy (AFM). Methods Conventional laboratory cell extraction methods, such as the pressing tissue cell method, cell impression preparation, and enzyme digestion centrifugal smear method, were used to prepare the thyroid in vivo tissue samples for observation under AFM. The cell dispersion and image definition of samples under the optical microscopy and AFM were compared to find the best sample preparation method. Results It was easy to find a single cell prepared by cell impression preparation when scanned by AFM , and there were few impurities, clear images and little interference. The pressing tissue cell method and enzyme digestion centrifugal smear method were not suitable for AFM. Conclusions The cell impression preparation method is the most suitable sample preparation method for AFM to observe the thyroid tissue cells. AFM can be feasibly used to scan the nanometer - grade structure of the thyroid cells, making it possible to research normal thyroid cells and thyroid tumor cells.
