摘要
目的观察缝隙连接阻滞剂heptanol对局部心肌缺血性室性心律失常发生率的影响,并探索可能的作用机制。方法结扎离体SD大鼠冠状动脉前降支,造成左心室前壁局部缺血。观察不同浓度heptanol对缺血所致室性心律失常发生率的影响。记录在0、缺血10、20及30rain各时间点动作电位复极90%时限(MAPD90)、心率、PR间期以及QT间期的变化。结果heptanol可以明显降低由于缺血引起的室性心动过速(室速)和心室颤动(室颤,缺血组45%;0.1mmol组10%;0.3mmol组0;0.5mmol组0;P〈0.05)。heptanol可以降低心率,延长PR间期、QT间期和MAPD90结论heptanol可以减少由于局部缺血引起的室速和室颤发生率,这种作用可能与其降低心肌电传导有关。
Objective To evaluate the effects of gap junction inhibitors heptanol on the electrophysiology and ventricular arrhythmias induced by regional ischemia of isolated SD rat hearts and to explore its possible mechanism. Methods The ischemia model was established by ligation of the left anterior descending coronary artery. The effects of heptanol on the occurrence of ventricular tachycardia and ventricular fibrillation were observed. Surface ECG, MAPD90, Heart rates, PR interval and QT interval were measured. The changes of parameters above were observed at 0 min,ischemic 10 min(Is 10 min) ,Is 20 min and Is 30 min. Results Heptanol could decrease the percentage of ventricular arrhythmia( ischemia control group ,45% ;0. 1 mmol/L group, 10; 0. 3 mmol/L group,0 ; 0. 5 mmol/L group,0 ; P 〈 0. 05 ), respectively. Heptanol could decrease heart rates, prolong PR interval, QT interval and MAPD90. Conclusion Heptanol could decrease the occurrence of ventricular arrhythmias induced by regional ischemia. That may be related to the slowing of electrical conduction.
