乳与乳制品中叶黄素的测定-高效液相色谱法

Determination of Lutein in milk and dairy products by high performance liquid chromatography

建立了乳与乳制品中叶黄素的测定方法。样品经皂化后,由乙醚提取,浓缩后经C30柱分离,高效液相色谱紫外检测器于445 nm处检测,外标法定量。叶黄素标准溶液在0.05～1.25 mg/L范围内呈线性相关(r=0.9997),不同基质中添加回收率为80.5%～91.3%,牛奶中叶黄素检出限为1μg/100g,奶粉、奶酪中叶黄素检出限为4μg/100g。试验证明本方法是一种准确测定乳与乳制品中叶黄素的方法。

The method of determination of lutein in milk and dairy products was developed.Samples were saponified and extracted by ethyl ether.Separated by C30 column,lutein was quantified by HPLC with ultraviolet detector at 445 nm.Lutein standard solution had a good linear correlation（r=0.9997） between 0.05～1.25 μg/mL,the recovery rate in different spiked samples range from 80.5% to 91.3%,with the LOD of 1 μg/100g in milk and 4 μg/100g in milk powder and cheese.The method is accurate and suitable for the quantification of lutein in milk and dairy products.
