ICP-MS研究中药炮制前后18种微量元素总量和溶出特性的变化 被引量:29

Effect of Processing on Content and Dissolution of 18 Trace Elements in Traditional Chinese Medicines by ICP-MS

摘要 目的应用电感耦合等离子体质谱(ICP-MS)技术对中药炮制前后18种微量元素的总量及溶出特性的改变进行考察。方法应用ICP-MS技术建立中药中18种元素的同时测定方法,并用该方法对黄芪、大黄、黄芩、何首乌、地黄等5种中药材炮制前后元素总量和水煎过程中溶出特性的改变进行比较和分析。结果测定方法中所有元素的线性相关系数(r)都大于0.999 0,检出限在0.000 1~0.068 9μg.L-1之间,重复性和稳定性的相对标准偏差(RSD)都小于10%,药材的加标回收率在70.89%~122.4%之间,水煎液的加标回收率在74.65%~130.0%之间,标准物质茶叶(GBW 07605)的测定值和其标定值之间有良好的吻合性。测定结果显示,炮制后,微量元素总量的变化因中药品种和炮制方法而异,但各元素的溶出率总体呈现上升的趋势。结论中药炮制可改变微量元素的总量和溶出特性。 OBJECTIVE To investigate content and dissolution feature changes of 18 trace elements in crude and processed traditional Chinese medicines(TCMs) by inductively coupled plasma mass spectrometry(ICP-MS).METHODS An ICP-MS method for simultaneous determination of 18 trace elements in TCMs was established,and the contents and dissolution features of 18 trace elements in 5 crude and processed TCMs,Radix Astragali,Radix et Rhizoma Rhei,Radix Scutellariae,Radix Polygoni Multiflori and Radix Rehmanniae,were compared and analyzed.RESULTS The validated method indicated that the correlative coefficients(r) for all elements were above 0.999 0.The limits of detections were in the range of from 0.000 1 to 0.068 9 μg·L^-1.The reproducibility and stability were satisfactory with all RSDs below 10%.The spiked recoveries for TCMs were between 70.89% and 122.4%.The spiked recoveries for their water extract solutions ranged from 74.65% to 130.0%,the determined values of tea reference material(GBW 07625) were in accordance with those certified.The changes of element contents depended on the TCM species and processing methods,while the dissolution rate of elements were generally increased after processing.CONCLUSION Processing TCM could change its contents and dissolution features of elements.
