摘要
以白燕2号为材料,研究了不同的外植体、培养基比例和外植体的放置方式对燕麦愈伤组织诱导的影响,以及最有效的消毒方法。试验结果表明,外植体最有效的消毒方法是采用75%酒精消毒1min后无菌水冲洗,再用30%的次氯酸钠溶液消毒40min(间隔20min换1次);诱导培养基中2,4-D最适浓度为3mg/L;燕麦种子是较理想的外植体,接种时将种子盾片朝下放置直接接触培养基,会得到更高更理想的出愈率。
This experiment was to select effective sterilization of explants and determine an optimun method of inducing oat callus by selecting suitable explants,2,4-D concentration in culture medium and placement of explants with oat cultivar "Baiyan 2".The results showed that the effective method of explant sterilization was sterilizing with 75% alcohol for 1 minute,rinsing with sterilized water,and then with 30% hypochlorite for 40 minutes (changing 30% hypochlorite every 20 minutes).The optimum concentration of 2,4-D in the induction medium was 3 mg/L.Oat seeds were suitable induction materials.Seed scutellum should be placed down to directly contact medium to obtain more desirable callus.
