摘要
建立测定头孢噻呋混悬注射液含量的RP-HPLC法。采用反相高效液相色谱,色谱柱为DiamonsilC18(4.6 mm×250 mm,5μm),流动相为乙腈-磷酸水(φ磷酸=0.1%,V乙腈∶V磷酸水=28∶72),流速1.0mL/min,检测波长292 nm。结果显示,测得3批头孢噻呋混悬注射液中头孢噻呋的含量分别为标示量的97.47%、98.02%和97.25%;头孢噻呋在0.0364~1.456μg内与峰面积线性关系良好(r=0.999 6),平均回收率分别为98.08%、98.95%、98.70%,RSD分别为1.10%、1.12%、0.94%。该方法专属性强,精密度好,准确可靠,可用于测定头孢噻呋混悬注射液中头孢噻呋的含量。
To establish an HPLC method for determination the ceftiofur content of the Ceftiofur Suspension Injection (CSI), the samples were separated on Diamonsil C18, (4.6 min × 250 mm,5um) column with the mobile phase of acetonitrile--phosphoric acid aqueous solution (φH3PO4 =0. 1%,28 : 72, v/v), flow rate 1.0 mL/min and the detection wavelength set at 292 nm. Results showed that ceftiofur content of three CSI were 97.47% , 98.02%, and 97.25% of the labelled quantitative, respectively. The linear range of ceftiofur was 0. 0364 - 1. 456 ug (r = 0. 999 6), the average recovery were 98.08%, 98.95%and 98.70%, and RSD were 1. 10%, 1. 12% and 0.94% (n=9), respectively. In conclusion, the established RP-HPLC method of ceftiofur quantitation was accurate and reliable,and can be used to evaluate the quality of Ceftiofur Suspension Injection.
