系统性红斑狼疮患者血浆一氧化氮及过氧化脂质的变化 被引量:3

摘要 目的检测SLE患者血浆中一氧化氮(NO)和过氧化脂质(LPO)水平,分析两者的关系。方法用生物还原法和比色法测定血浆中NO-2/NO-3总浓度,以此代表血浆中NO水平,用TBA显色法测定LPO浓度。34例SLE患者中23例为初次治疗的活动期SLE,其中15例口服强的松(1mg/kg)治疗,11例为稳定期SLE,30例健康人为对照组。结果活动期SLE血浆NO水平(5131±3023)μmol/L高于正常对照组(3656±2150)μmol/L,15例活动期SLE治疗一周后血浆NO水平由(5926±2979)μmol/L降至(4650±2421)μmol/L,且活动积分下降。活动期SLE血浆LPO水平(569±222)μmol/L显著高于对照组(397±074)μmol/L,血浆NO水平与LPO水平无直线相关性。结论血浆NO水平可作为SLE活动指标之一,糖皮质激素对NO合成有抑制作用,活动期SLE体内存在氧化损伤。 Objective To determine the levels of plasma nitric oxide (NO) and lipid peroxides (LPO) of patients with systemic lupus erythematosus (SLE),and analyse the relationship between them.And to observe the NO level change of active SLE patients before and after treatment with prednisone (1 mg/kg).Methods Plasma NO - 2/NO - 3 (stable end production of NO in plasma) levels of 34 SLE patients (23 active SLE patients,11 stable SLE patients) and 30 healthy subjects were determined by organism reduction and spectrophotometric assays.The levels obtained represented NO levels in plasma.Plasma LPO levels were determined by the thiobarbituric acid (TBA) reaction.Results The level of plasma NO of active SLE group was higher than that of healthy group (51 31±30 23)μmol/L,(36 56±21 50)μmol/L.The level of plasma NO after one week treatment declined from (59 26±29 79)μmol/L to (46 50±24 21)μmol/L with the SLE DAI dropped average 3 31 score (0~18 score).The level of plasma LPO of active SLE group was higher than that of healthy group (5 69±2 22)μmol/L,(3 97±0 74)μmol/L.Although the levels of both plasma NO and LPO were higher in SLE,no obvious relationship was observed between them.Conclusion Results show that the level change of plasma NO is consistent with the results of other authors' animal studies.The level of plasma NO can be considered one of the standards of SLE activity.Glucocorticoids can inhibit NO synthesis.There is oxidative injury in active SLE.The relationship between oxidative injury and NO is not sure.
