Serum proteomic analysis in spontaneously hypertensive rats

摘要 目的探讨自发性高血压大鼠(SHR)蛋白质组学的发病机制。方法以SHR10只为模型组,Wistar Kyoto大鼠10只为正常组,取主动脉血,低温离心,分离血清,每组随机各取5份血清样品,用双向凝胶电泳、基质辅助激光解析电离飞行时间质谱测定肽质量指纹图谱,数据库检索鉴定各肽段对应的蛋白质,获取血清差异表达蛋白质的信息。结果 2组均获得重复性好的血清蛋白质双向凝胶电泳图谱。模型组和正常组凝胶上的平均蛋白点分别为(527±34)个、(476±30)个。通过分析蛋白质点局部图谱,获得23个差异表达蛋白点。与正常组比较,模型组4个点高表达,17个点低表达,2个点缺失。23个差异点经鉴定可以确认的蛋白质点有:线粒体膜蛋白、锌指蛋白、Rho GTPase、T细胞受体、视黄醇结合蛋白、假想蛋白。结论线粒体膜蛋白、锌指蛋白、T细胞受体、假想蛋白在SHR低表达;Rho GTPase、视黄醇结合蛋白在SHR高表达,推测这些蛋白可能与高血压的发生、发展密切相关。 Objective To investigate the pathogenesis of hypertension from view-point of serum proteomics in SHR. Methods Ten SHR were selected as model group,10 Wistar Kyoto rats were selected as normal group. Aortic blood was taken and centrifugalized at low temperature. Five serum samples were taken from each group to detect the peptide mass finger print by two-dimensional gel electrophoresis and MALDI-TOF-MS. Database search was used to identify the proteins corresponding to the peptides and obtain the information of the different expression proteins. Resuits The two-dimensional gel electrophoreto grams of serum proteins were successfully build for normal,model and Chinese medicine groups. The average protein points of model and normal groups were 527±34 and 476±30. Compared with normal group,there were 23 significantly different points in model group,of which 4 points were in high expression, 17 points were in low expression and 2 points were lost in SHR. Six different protein points were identified:mitochondrial membrane protein, zinc finger protein, Rho GTPase, T-cell receptor, retinol binding protein, hypothetical protein. Conclusion Mitochondrial membrane protein,zinc finger protein, T-cell receptor and hypothetical protein are in low expression in SHR; while Rho GTPase and retinol binding protein are in high expression. It is inferred that these proteins may be closely related to the occurrence and development of hypertension.
