玉葵清对AGEs干预的人肾小球系膜细胞CTGF表达的影响 被引量:2

Effect of Yukuiqing on expression of connective tissue growth factor in human real mesangial cells incubated with AGEs

摘要 目的:探讨玉葵清对糖基化终产物(AGEs)干预的人肾小球系膜细胞(HRMC)结缔组织生长因子(CTGF)mRNA及其蛋白表达的影响。方法:用中药血清药理学方法制备玉葵清含药血清(YKQ);用同一浓度YKQ(1.25%)和AGEs(200μg/mL)干预HRMC不同的时间(0、8、16、24、48、72h)以及用不同浓度的YKQ(0.313%、0.625%、1.25%)和AGEs(200μg/mL)干预HRMC48h,以无血清培养基(DMEM)和相应浓度的鼠血清(RS)为对照,用RT-PCR法和Western Blotting分别检测CTGF mRNA和蛋白的表达水平。结果:与0h相比,YKQ呈时间依赖减轻AGEs干预的HRMC CTGF mRNA和蛋白的表达(P<0.05)以及YKQ浓度在0.313%、0.625%和1.25%时,较对照组显著减轻CTGF mRNA和蛋白的表达(P<0.05)。结论:玉葵清能抑制AGEs干预的HRMC CTGF mRNA和蛋白的表达,对糖尿病肾病起一定的防治作用,此作用可能是其防治糖尿病肾病的机制之一。 Objective:To investigate the effects of Yukuiqing (YKQ) on the expression of connective tissue growth factor (CTGF) in cultured human renal mesangial cells (HRMC) incubated with AGEs. Methods:The HRMCs were incubated with AGEs (200 μg/mL) and 1.25% YKQ which was prepared by Chinese herbal medicine serum pharmacological approach for 0,8,16,24,48 and 72h or different concentrations (0.313%,0.625% and 1.25%) of YKQ for 48h,respectively,which were incubated with DMEM and rat serum (RS) as the control. CTGF mRNA and protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western Blotting respectively. Results:In Yukuiqing group,the expression of CTGF mRNA and protein decreased significantly compared with control groups (P0.05) and this decrease was in time dependent manner. The same result appeared in Yukuiqing group (0.313%,0.625%,1.25%) compared with control groups (P0.05). Conclusion:YKQ attenuated the expression of CTGF mRNA and protein in HRMCs incubated with AGEs. These results indicated that Yukuiqing might play the role of reno-protection via suppression of CTGF induced by AGEs.
