摘要 目的:研究GnRHⅡ对子宫内膜异位症(内异症)患者离体培养的异位及在位子宫内膜间质细胞凋亡的影响。方法:体外原代培养人内异症异位及在位子宫内膜间质细胞,用不同浓度的GnRHⅡ(0、10-10、10-8、10-6mol/L)干预,采用Hoechst染色和流式细胞术检测离体培养子宫内膜间质细胞的凋亡发生情况。结果:GnRHⅡ可诱导离体培养的在位或异位子宫内膜间质细胞的凋亡,随着浓度增加,差异有统计学意义(P<0.05),呈剂量依赖性;且对异位内膜间质细胞的促凋亡作用明显强于在位者(P<0.05)。结论:外源性GnRHⅡ可明显促进人内异症异位及在位子宫内膜间质细胞的凋亡,为开发内异症促凋亡治疗方面的新药提供了实验依据。 Objective:To investigate the effect of GnRH Ⅱ on the cell apoptosis of ectopic and eutopic endometrial stromal cells from women with endometriosis in vitro. Methods: The ectopic and eutopic endometrial stromal cells were isolated and cultured in vitro, then exposed to different concentration of GnRH Ⅱ(0,10^-10, 10^-8 , 10^-6mol/L). The cell apoptosis of ectopic and eutopic endometrial stromal cells were measured by Hoechst staining and flow cy- tometry test. Results:GnRH Ⅱ can increase the apoptotic rate of both ectopic and eutopic cells in a dose-dependent manner(P〈0.05 ) , and the apoptotic rate of ectopic stromal cells was obviously higher than that of the eutopic cells (P〈0.05). Conclusion: GnRH Ⅱ can significantly enhance the apoptosis of ectopic and eutopic endometrial stromal cells from women with cndometriosis,which will provide the evidence for researching endometriosis drug treatment.
