摘要
目的:揭示1例银屑病患者骨髓间充质干细胞体外自发向血管内皮细胞分化。方法:采用密度梯度离心法分离骨髓单一核细胞,通过贴壁法培养骨髓间充质干细胞,经全量换液和2次半量换液后(即培养9 d)流式细胞术鉴定正常对照细胞免疫表型,再培养2d,采用流式细胞术和Dil-ac-LDL、FITC-UEA-1双荧光染色法,分别对患者的细胞和正常对照细胞进行内皮细胞鉴定。结果:患者细胞在培养11 d后大部分分化为血管内皮细胞,而正常对照则仍为较纯的间充质干细胞。结论:该患者骨髓间充质干细胞体外培养自发向血管内皮细胞分化,这一现象表明银屑病患者骨髓间充质干细胞向血管内皮细胞分化过程中可能存在异常,其间充质干细胞有易于向血管内皮细胞分化的倾向。
Objective: To reveal that a case of the vascular endothelial cells spontaneously differentiated in vitro from bone marrow derived mesenchymal stem cells in patient with psoriasis by identifying the biological character (including morphology, immunopheno- type and the uptaking Dilac-LDL characteristic). Methods:Separate bone marrow mononuclear cells by density gradient centrifuga- tion. Bone marrow derived mesenchymal stem cells were cultured with the method of adherence. Cells of control at cultured for 9 days were collected. Flow cytometry analysis was used to examine the immunophenotype. The identification of patients" and controls" endothelial cells was separately performed. Cells cultured for 11 days examined by flow cytometry analysis and Dil-ac-LDI./FITC-UEA-1 dual-fluorescence staining. Results: Cells from patient cultured for 11 days mostly differentiated into vascular endothelial cells. But cells from controls were mostly bone marrow derived mesenchymal stem cells. Conclusions: Bone marrow derived mesenehymal stem cells in patient with psoriasis spontaneously differentiated in vitro into the vascular endothelial cells. This phenomenon shows that bone marrow derived mesenchymal stem cells in patient with psoriasis may be abnormal during the differentiation to vascular endothelial cells. Bone marrow derived mesenchymal stem cells in patient have the tendency of differentiating to vascular endothelial cells.
