摘要
目的探讨中国北方汉族人中HLA-A、B、DRB1等位基因与肺癌遗传易感性间的关系。方法采用测序分型技术(sequence-based typing,SBT)技术,对无血缘关系的籍贯为中国北方的140名肺癌患者及483名健康志愿者的HLA-A、B、DRB1基因进行检测。arlequin软件(ver2.000)及SPSS软件进行统计分析。结果 A*2601、B*1518、B*3802、DRB1*0401、DRB1*0402、DRB1*1201在肺癌病人中频率显著性高于正常对照,P值分别为0.021、0.001、0.015、0.021、0.010和0.046,OR值分别为3.513、3.842、2.715、3.512、13.986、1.828;HLA-DRB1*1001、DRB1*1302在肺癌病人中频率显著性低于正常对照,P值分别为0.017和0.014,OR值分别为0.135和0.122。单倍型HLA-A*0207-B*4601-DRB1*0901、HLA-A*0206-B*5101在肺癌病人中频率显著性高于正常对照,P值分别为0.034和0.006,OR值分别为2.348和3.969;单倍型HLA-A*1101-DRB1*1501在肺癌病人中频率显著性低于正常对照,P值为0.026,OR为0.146,另外单倍型HLA-A*0101-B*3701在正常对照中频率为0.02070但在肺癌病人中未检出。结论在中国北方人中HLA-A*2601、B*1518、B*3802、DRB1*0401、DRB1*0402、DRB1*1201和单倍型HLA-A*0207-B*4601-DRB1*0901、HLA-A*0206-B*5101与肺癌发病正相关,HLA-DRB1*1001、DRB1*1302和单倍型HLA-A*1101-DRB1*1501、HLA-A*0101-B*3701与肺癌发病负相关。
The primary function of HLA system is to regulate the immune response.HLA system is associated with many diseases,due to it plays important roles in the immune response and high polymorphism.In the present study,we examined the polymorphisms of HLA-A,B and DRB1 alleles in 140 unrelated patients with lung carcinoma and 438 unrelated normal controls of Han nationality from North China using sequence-based typing(SBT) and polymerase chain reaction with sequence-specific primer(PCRSSP).We found that the frequencies of HLA-A*2601,B*1518,B*3802,DRB1*0401,DRB1*0402 and DRB1*1201 were higher in the lung carcinoma group than those in the normal control group,with P values were respectively 0.021,0.001,0.015,0.021,0.010 and 0.046,while OR values were respectively 3.513,3.842,2.715,3.512,13.986 and 1.828.The frequencies of HLA-DRB1*1001 and HLA-DRB1*1302 were lower in the lung carcinoma group than those in the normal control group,with P values were respectively 0.017 and 0.014,while OR values were respectively 0.135 and 0.122.The frequencies of haplotype HLA-A*0207-B*4601 DRB1*0901 and HLA-A*0206-B*5101 were higher in lung carcinoma group than those in normal control group,whose P values were respectively 0.034 and 0.006,while OR values were respectively 2.348 and 3.969.The frequency of haplotype HLA-A*1101 DRB1*1501 was lower in that lung carcinoma group than in normal control group,with P value was 0.026 and OR was 0.146.The frequency of haplotype HLA-A*0101-B*3701 in normal control group was 0.02070 but not found in lung carcinoma group.Our results suggest that the frequencies of some HLA alleles and haplotypes do related with the susceptibility to lung carcinoma of Han nationality from North China.
