摘要
为进一步探讨济南假单胞菌制剂(PJA)抗瘤作用机制,将PJA作用于人高转移肺癌细胞PG,采用MTT、放免、流式细胞术、粘附实验、运动侵袭实验等多种手段进行了检测。显示PJA有显著的抑瘤作用,并能影响PG细胞周期的时相分布,使S期减小。其对PG细胞内cAMP无明显影响,但能降低cGMP水平,提高cAMP/cGMP比值,同时降低PG细胞对胞外基质的粘附性及侵袭、运动能力,从而逆转肿瘤细胞的恶性表型。提示,PJA作为一种新型抗癌药物具有较高的临床应用价值。
Pseudomonas Jinanensis Agent(PJA) is a kind of new anticancer drug. In order to further elucidate the anticancer mechanism of PJA, highly metastatic human lung cancer cells(PG) were treated with PJA in vitro . MTT assay, radioimmune assay, flow cytometry(FCM), and invasion assay were used. The results showed that PJA could inhibit the proliferation of PG, influenced the distribution of PG cells cycle and S phase was reduced. Although PJA didn′t change the level of cAMP in PG cells, it reduced the level of cGMP and increased the cAMP/cGMP ratio. On the other hand, PJA also decreased PG cells adhesive ratio to laminin substrate and decreased the ability of invasion or migration. These data demonstrated that PJA is a kind of effective anticancer drug.
