p16与c-myc基因表达产物在肝细胞癌及癌旁组织中的表达及临床意义 被引量:3

摘要 目的:探讨p16、c-myc蛋白与肝细胞癌(hepatocelularcarcinoma,HCC)的发生、发展及恶性程度的关系。方法:应用免疫组织化学法检测了69例HCC和63例癌旁组织p16及c-myc蛋白的表达,并结合临床资料进行分析。结果:p16蛋白主要分布在胞浆,少数细胞核内也可见阳性显色。c-myc蛋白主要分布在胞核。p16蛋白在肝癌组织及癌旁组织中的阳性率分别为26.5%(18/69)和92%(58/63),两者比较差异显著(P<0.01)。69例HCC标本,Ⅰ级、Ⅱ级、Ⅲ级中p16蛋白的检出率分别为52.6%(10/19),23.3%(7/30),5%(1/20),p16蛋白在高分化HCC中的表达率明显高于低分化HCC组(P<0.05)。c-myc蛋白在癌组织及癌旁组织中的阳性率分别为85.5%(59/69)和39.7%(25/63),两者比较差异显著(P<0.05)。结论:抑癌基因p16和c-myc基因的表达和分布与HCC发生、发展及临床预后有密切的关系。 Objective:To study the relationship between the expression of P16,the c-myc protein and the development and malignancy in primary hepatocellular carcinoma (HCC).Methods:P16 and c-myc protein expressions were detected in 69 specimen of HCC and in 63 specimen of pericarcinoma tissues by the method of immunohistochemistry.The data was analyzed with the clinical meterials.Results:The positive immunocytochemical staning of P16 protein was located mainly in cytoplasm and the positive staining was also identified in nucleolus of a small number of cells.The c-myc protein was detected Within the nucleouls region.The positive rate of P16 protein expression was 26.5% (18/69) in HCC samples and 92% (58/63) in Pericarcinoma tissues,the difference of the rate between the two groups was significant (P<0.01).In 69 specimen of HCC,the positive expression rate of P16 protein among samples of grade Ⅰ,Ⅱ and Ⅲ were 52.6%(10/19),23.3%(7/30) and 5%(1/20),respectively.P16 protein expression rate of highly differentiated HCC was significantly higher than that in lower differentiated ones (P<0.05).The positive rate of c-myc protein expression was 85.5% (59/69) in HCC samples and 39.7%(25/63) in pericarcinoma tissues with statistical significance (P<0.05).The c-myc protein expression in HCC was negatively interrelated with cell differentiation.Conclusion:The expression and distribution of the p16 and c-myc genes were closely related with the generation,development and clinical prognosis of HCC.The detection of p16 and c-myc may have important significance to clinical treatment for HCC.
