摘要
目的:探讨骨形成蛋白(BMP)与细胞凋亡的关系及意义。方法:利用脂质体转染方法将BMP2真核表达载体pBK-B2导入NIH3T3细胞,通过G-418筛选获得阳性细胞克隆。细胞原位杂交和免疫组化检测BMP2基因的稳定转染及表达,并利用TUNEL法观察细胞凋亡情况。结果:BMP2基因成功导入NIH3T3细胞并得到表达。转染细胞的凋亡细胞阳性率与未转染细胞相比有显著性差异。结论:BMP2能够诱导细胞凋亡,且这种作用与细胞所处的生长分化状态密切相关。
Aim: To elucidate the relationship between Bone Morphogenetic Protein (BMP) and cell apoptosis. Methods: Eukaryonic expression vector (pBK-B2) for BMP-2 was transduced into NIH3T3 cells by using LipofectAMINE. Positive cell clones were selected with G-418. The stably transfection and expression of BMP2 in the NIH3T3 cells were determined by in situ hybridization and immunohistochemistry methods. Cell apoptosis was also observed by TUNEL methods. Results: BMP2 gene was successfully transfected and expressed in NIH3T3 cells. There was significant difference in the rate of apoptosis between transfected cells and non-transfected cells. Conclusions: BMP2 can induce cell apoptosis and this effects are closely related to the state of cells.
