摘要
用重量法对水蕨[Ceratopteris thalictroides (L.) Brongn.]愈伤组织生长进行了初步探索。结果表明,以MS培养基为基本培养基附加不同外源激素均可诱导产生水蕨的愈伤组织,但不同浓度植物激素对愈伤组织生长影响不同;生长素NAA对水蕨外植体的脱分化起促进作用;2,4-D却抑制愈伤组织的形成;细胞分裂素6-BA能与NAA组合促进愈伤组织的诱导。在光暗交替条件下,水蕨愈伤组织的最佳诱导和增殖条件是MS+6-BA1.0mg/L+NAA0.5mg/L。在此条件下,愈伤组织继代培养20d后,平均增殖率达1122.70%。
Gravimetry to measure the callus induction of Ceratopteris thalictroides was preliminary explored in this paper. The results showed that the callus of Ceratopteris thalictroides could be induced on MS medium supplemented with different kinds of hormones. The frequency of callus induction was different when medium supplemented with different types and different concentrations of hormones. NAA could promote to dedifferentiation,but 2,4-D could restraint the callus formation,and 6-BA mixed with NAA both promoted the induction. The optimized condition for induction and multiplication was MS + 6-BA1.0 mg/L + NAA0.5 mg/L in darkness and light in turn (12 hours illumination every day). On this condition,after it had been subculturing for 20 days,the average multiplication rates could be up to 1 122.70%.
